Spectroscopy
We investigate the haem-copper oxidases after biochemical purification by means of different biophysical techniques, mainly using optical and Fourier-transform infrared spectroscopy (FTIR-spectroscopy). Our major goal is the identification of amino acids involved in proton pumping and the investigation of their exact role in catalysis. For the assignment of spectroscopic signals we combine gene technological and biochemical methods (i. e. site-directed mutagenesis, in-vivo isotopic labelling).
